CpG-ìåòèëèðîâàíèå — ýïèãåíåòè÷åñêàÿ ìîäèôèêàöèÿ, âàæíàÿ äëÿ ýìáðèîíàëüíîãî ðàçâèòèÿ, èìïðèíòèíãà è èíàêòèâàöèè Õ-õðîìîñîìû. Èññëåäîâàíèÿ íà ìûøàõ ïîêàçàëè, ÷òî ìåòèëèðîâàíèå ÄÍÊ òðåáóåòñÿ äëÿ ðàçâèòèÿ ìëåêîïèòàþùèõ. Ãåí DNMT3L êîäèðóåò ÿäåðíûé áåëîê, ïîõîæèé íà ÄÍÊ-ìåòèëòðàíñôåðàçó. Ýòîò áåëîê íå ôóíêöèîíèðóåò êàê ÄÍÊ-ìåòèëòðàíñôåðàçà, òàê êàê îí íå ñîäåðæèò àìèíîêèñëîòíûõ îñòàòêîâ, íåîáõîäèìûõ äëÿ àêòèâíîñòè ìåòèëòðàíñôåðàçû. Òåì íå ìåíåå, ýòîò áåëîê ñòèìóëèðóåò de novo[àíãë.] ìåòèëèðîâàíèå ÄÍÊ-öèòîçèí-ìåòèëòðàíñôåðàçû 3-, è ñ÷èòàåòñÿ, ÷òî îí íåîáõîäèì äëÿ ñîçäàíèÿ ìàòåðèíñêèõ ãåíîìíûõ èìïðèíòîâ. Ýòîò áåëîê òàêæå ÿâëÿåòñÿ ïîñðåäíèêîì ðåïðåññèé òðàíñêðèïöèè ïóò¸ì âçàèìîäåéñòâèÿ ñ ãèñòîíäåàöåòèëàçîé 1 (HDAC1). Àëüòåðíàòèâíûé ñïëàéñèíã äà¸ò äâà âàðèàíòà òðàíñêðèïòîâ. Áûë îïèñàí äîïîëíèòåëüíûé âàðèàíò ñïëàéñèíãà, íî åãî áèîëîãè÷åñêîå äåéñòâèå íå áûëî îïðåäåëåíî[2].
